This important new book provides the fundamental understanding of the peptide and protein drug delivery systems with a special focus on their nanotechnology applications. Addressing an increasing interest in peptide and protein drug delivery systems in both academic and industrial circles worldwide, this book fills the need for a comprehensive review and assessment of conventional and nonconventional routes of administration.

Drug delivery is a term that refers to the delivery of pharmaceutical compound
to humans or animals. The most common methods of delivery
include the preferred noninvasive oral (through the mouth), nasal, inhalation,
and rectal routes. Many medications, however, cannot be delivered
using these routes because they might be susceptible to degradation or
not incorporated efficiently. For this reason any protein and peptide drugs
have to be delivered by injection.
Human cellular and tissue membrane always resist the entry of foreign
material (medicine). Selection of efficient drug delivery system allows efficient
delivery for medicine into the body. Due to high stability, high carrier
capacity, feasibility of incorporation of both hydrophilic and hydrophobic
substances, and feasibility of variable routes of administration, including
oral application and inhalation, nanoparticles are always preferred. Small
peptides can easily transport drug, protein to a specific region in the cell,
including the nucleus, mitochondria, endoplasmic reticulum (ER), chloroplast,
apoplast, peroxisome and plasma membrane. Some target peptides
are cleaved from the protein by signal peptidases after the proteins are
transported. For the specified targeting of the nanoparticle at particular
site, its bioconjugation with peptide is essential. This type of bioconjugation
is widely used to deliver drugs, proteins, DNA/RNA, viruses, and
vaccines to the human body to improve the quality of human health.
This book provides the fundamental understanding of drug delivery
systems of peptide and proteins with a special focus on their nanotechnology
applications. Since this book was first conceived, there has been
increasing research efforts dedicated to peptide and protein drug delivery
systems in both academics and industrial laboratories worldwide. This
book was written to fulfill a need for a comprehensive review and assessment
of conventional and nonconventional routes of administration.
To this end I have organized Chapter 1 so that it gives fundamental
and comprehensive knowledge regarding the synthesis, stability, degradation,
and physicochemical aspects of proteins and peptides. In addition this part also provides the comprehensive information related with barriers
to peptide and protein delivery and their respective formulation and
delivery strategies.
Chapter 2 discusses the pharmacokinetic consideration, methods of
preparation, design and synthesis of peptides, structure function relationship,
peptide polymer conjugate, peptide-encapsulating nanoparticles,
biomedical applications of peptide polymer conjugate, and nonpeptide
targeting.
Chapter 3 discusses the CPP and CTP in drug delivery and cell targeting.
This book fulfill my expectations if it creates interest in the area of
peptide and protein drug delivery and provides a framework for the pharmaceutical
or biotechnological scientist to formulate a strategy to deliver
peptide and protein drugs to their site of action.
I am highly thankful to my caring and lovable parents, Mr. R. L. Bhatia
(Father), Vinod Bhatia (Mother), my dearest brother, Sanjay Bhatia, and
my sweet sister-in-law, Aastha Popli Bhatia, for their caring, valuable
suggestions and timely input. This book would not have seen the light of
the day without the moral support and patience of my family. In addition
Ashish Kumar, Sandy Jones Sickels, and Rakesh Kumar, all of Apple
Academic Press, must be praised for their active work and support for our
effort. We are sure that readers of this book—students, researchers, and
industrialists—will find it interesting and useful. The publication of this
book would not have been possible without the valuable work of earlier
researchers.
